Sigil Question

Sorry if this is a dumb question but are the positions important? I would assume yes. What are the benefits of putting them "where they're supposed to be" rather than just having them at all and slotting them anywhere? Should they all be slotted 135 246? Does it show the benefits in game somewhere that I just don't know where to find?
